ABSTRACT:This study was included isolation and identification of bacterial isolates for growth promoter production (Indol Acetic Acid) by application of local culture and evaluate its efficiency. According to this perpoute bacterial isolates were and examined for their ability to indol production, and then serial labrotory experiments were converted to examine ability of selective isolates for indol production. The local culture used included dry powder of legume, bean, soybean, seed and milk powder. Its also was tested the effect of edition of supplements to the culture such as treptophan, N. broth, glucose, N and P on indol production optimum condition was included such as pH speed of shaking, Inoculums volume, incubation time, temperature, periods of storage to increase production efficiency.This study have the following results: 18 isolates were obtain capable for them indol production (30 from total isolates) 14 isolates were nodulan bacteria .8 isolates which have high efficancy in indol production were tested six isolates tended to Rhizopium and two isolate to Pseudomonus in next screening used culture with special condition, two isolates Pssp2S, Rsp8RA were selected to be used in the following epperinets. When we used 10% of local culture prepared from dry powder of legumes, bean, soybean seed, milk powder led to increased of indol production significantly ranged from (30- 50%). Local culture supplements with peptone, NB, glucose, N and P increased the ability of isolates to indol production (40-65%) significantly.The results of optimum condition study showed the best indol production a chaired with R. sp8RA isolat with pH 7.0 ( 39.41 mg IAA / ml and the best shaking speed at 200/ min without aeration of media 42.2 mg IAA/ ml, the best interaction between the inoculation volume with incubation time with 3 ml/ 100 ml of media at 24 hr of incubation (42.8 mg IAA/ ml) in microbial population 8.41 Log cfu/ ml. Effect of temperature and period of filtrated isolates storage on speed of seed germination increased efficacy filtrated isolates that stored at 4C and 25C for one day and that stored at 4 ◌ْC for 45 days. When it compared with treatment filtrated storage at 25 ◌ْC for 45 days and treatment of industrial indol.The filtrated isolates achieved in commonly increase in speed and percentage of germination was reached 10 – 20 % in temporal differences 3 – 5 day respectively when compare with control treatment.

Afield experiment was carried out in Education college for women AL-Anbar University during the winter season 2009 – 2010 . the objective was to study the response of three wheat genotypes ( Austuraly S.D.30 , Abu grab3 , Sham6 ) for priming by three concentration of Mn (0 , 5,10) ppm – Mn and its effects on Germination , Growth , chlorophyll a and b concentration . Genotype wheat seeds were soaked in water and in different concentration of Mn (0 , 5 , 10) ppm for 12 hours then dried back and storage in dark at small plastic pots in the incubator at 40 for week. Afield experiment was carrid out according to split / plot system with randomized complete block design (R.C.B.D) . A total of 27 treatment with three replication , the area of each experiment unite was (2 x 2.5) m2. seeds was sown in rows in ten rows for unit treatment.A statistical analysis was done for data and treatment means were compared by least significant different (L.S.D) at 0.05 significants level. The Abu Graib3 genotype was showed asignificant effect in speed germination and leaf area 91.66 % and 25.84 cm2 respectivlly .The Sham6 genotype showed asignificant effect in Spike wight and chlorophyll a 3.66 gm 2.05 mg / gm respectivlly The genotypes had no significant effect on Spike length , chlorophyll b and biological yield . The concentration of Mn-seed Priming had asignificant effect on some studies characters .the concentration 10 ppm-Mn was the superior in speed germination , leaf area, chlorophyll a and biological yield83.22 %, 28.51 cm2 , 2.40 ml / gm, 1222 kg / h respectivlly. The priming treatment was showed no significant effect of spike length , spike weight , chlorophyll b . The interaction effect of Abu graib3 genotype and concentration 10 ppm- Mn was superior in spreed germination 93.67 %.The interaction effect austuraly S.D.30 and concentration 10 ppm-Mn was showed asignificant effect in leaf area 29.18 cm2. There was asignificant interaction between Sham6 and concentration 10 ppm-mn on spike weight , chlorophyll a , chlorophyll b and biological yield 3.80gm, 2.50 ml/gm, 1.55 ml/gm, 1300 kg / h.There was no significant interaction between wheat genotype and seed priming concentration on spike length.

The present study included the collection of 169 samples from various clinical and environmental sources for investigation of existence of Pseudomonas aeruginosa in those sources, the clinical samples collected from some hospitals in Baghdad city and included 126 samples of wound, burns, cystic fibrosis, ear infections and urinary tract infections (35, 52,4, 22 and 13) samples respectively, while environmental samples were distributed by 25 samples from soil, and 18 samples from wastewater and some swamps. Samples were identified based on the morphological and microscopical characteristics of the colonies when they were culturing in a number of culture media as well as biochemical tests. The molecular detection was used to confirm diagnostic test for the isolates based on the 16SrRNA gene, as a detection gene, which has specific sequences for DNA of P.aeruginosa bacteria.The results of molecular detection showed that 51 isolates belong to target bacteria were distributed as 20 of burns, 9 isolates of wounds, 7 isolates of ear infection, 4 isolates of cystic fibrosis, 2 isolates of urinary tract infection, 5 isolates of soil and 4 isolates of water. The results of phenotypic detection of some virulence factors showed that 49 isolates with percent (95%) produced beta-hemolysis, while 46 isolates (90%) showed variance in the strength of pycocyanin production with a absorbance ranging from (0 to 0.52) at 690 nm, and 47 (92%) of isolates were producing protease enzyme, 44 (86%) of the isolates showed a differences in their ability to biofilm formation. As a result of the increased resistance of P.aeruginosa bacteria towards antibiotics, it is became a problem in the treatment of the infectious diseases caused by this bacteria therefore, the current study focused on the use of fresh garlic extract towards of some virulence factors because it being easy and commonly used. The results of the statistical analysis showed a significant difference when P-value (<0.05) in the production of some virulence factors, the isolates lost their ability to form biofilm, and decreased their ability to produce protease enzyme, and pyocyanin pigment, while did not affect the production of hemolycin.

Result of applied biological experiment showed an increased treatment of two isolates mixture Rsp8RA , PssP2S with their filterated achaived inclues in : A-Characters of vegetative group that achaived high avarge for branching ( 4.37 ) height of plant ( 81.2 cfu/ ml and leaf area 78.4cm ) ,persente of chlorophyll ( 39.6% ), dry weight (51.2 g) Nitrogen and Phosphours yield in plant ( 491, 1020 mg ) respectively . B- Root characters were achaived high avarge the number of noduls (56.3 noduli) . weight of root group ( 10.4 gm ). c- Soil characters after sowing were achaived high average of nitrogen and phosphorus in soil (166 – 18.4 mg / kg soil) respectively and high microbial population in soil ( 7.82 Log cfu/ ml ).
